HORMONES AND FREE RADICALS
Aging is associated with a decline in hormones and an increase in free radicals that begins relatively early in life. Perturbations in growth hormone homeostasis occur first and can be detected clinically during the thirties. This change initiates a progressive cascade of general endocrine decline with each passing decade. Not coincidentally, there is a massive oxidative shift in the human plasma thiol/disulfide redox state between the 3rd and 10th decades of life. Because these changes in endocrinology and metabolism are correlated with progressive senescence, they are considered to be contributing factors to loss of health and vitality during aging. Accordingly, hormone replacement therapy (HRT) and antioxidant supplements are commonly used to compensate for them. However, these interventions in aging are often used as if hormone loss and free radical accumulation are independent events. In fact, they are functionally linked, i.e. hormonal decline contributes to loss of anti-oxidants and increased accumulation of specific free radical species. Also, there is often little consideration given to the type of free radical being targeted. Rather there tends to be a "shotgun" approach to the use of vitamins and other anti-oxidant supplements so as to rid the body of free radicals. This approach is flawed because like hormones which are essential to the body in certain amounts, at certain times and in association with other molecules, free radicals or their derivatives contribute to important physiological functions ranging from regulation of vascular tone to enhancement of signal transduction from various membrane receptors. Thus, flooding the body with antioxidants or over-expressing anti-oxidative enzymes may be just as detrimental as excessive HRT. To ensure ordered endocrine homeostasis and redox-mediated signaling, healthy life requires delicately balanced and appropriate levels of hormones, free radicals and radical-derived reactive oxygen species (ROS). Thus, HRT and anti-oxidant supplements should be used as integrated approaches so as to derive maximum efficacy and safety from their use.

With this concept in mind, the international Society for Applied Research in Aging (SARA) has scheduled a teaching seminar on hormones and free radicals that will include discussion of how they are interactive, how they mutually contribute to senescence and how physicians may use replacement therapy safely and effectively. Discussion of indirect GHRT using sermorelin (GRF1-29) in compliance with federal laws and good medical practice will be an important topic of interest since it opposes the primary locus of age-related endocrine decline. SARA members include anti-aging practitioners who are devoted to the ethical and professional advancement of their field, and who employ evidence-based interventions in aging as the central focus of their practice.
